Results The IS anti-tumor effect was dependent on both its N-alkyl chain length and anion, whereby C(16)MImCl proved to be more effective in combination for inhibiting cell proliferation and cell-cell adhesion, outperforming the methylated C(16)M(2)ImCl derivative and, most importantly, the gold standard-cisplatin. In addition, C(16)MImCl had little effect on keratinocytes and more pronounced effects on more aggressive tumor cells. It also exhibited similar effects on inducing cell death when compared to Cisplatin. This compound spread to a greater area of the tumor sphere and produced an enhanced number of apoptotic and necrotic cells in the tumor cell line, demonstrating only a small rise in the healthy cells. Conclusion These data indicate that the effect of C(16)MlmCl on OSCC is promising, as it is selective for cancer cells.

BACKGROUND The cys-loop ligand-gated ion channels, including the glutamate-gated chloride channel (GluCl) and GABA-gated chloride channel (Rdl) are important targets for drugs and pesticides. Next, we used CRISPR-Cas9 to introduce I321T, G314D and G326E in the orthologous Drosophila GluCl. Genome modified flies expressing GluCl I321T were threefold less susceptible to abamectin, while CRISPRed GluCl G314D and G326E flies were lethal. Last, functional analysis in Xenopus oocytes revealed that the I321T mutation might reduce GluCl3 sensitivity to abamectin, but also suggested that all three T. urticae Rdls are affected by abamectin. CONCLUSION Three different techniques were used to characterize the role of I321T in GluCl3 in abamectin resistance and, combining all results, our analysis suggests that the I321T mutation has a complex role in abamectin resistance. Given the reported subtle effect, additional synergistic factors in resistance warrant more investigation.

Under the optimum flocculation conditions (dosage = 16 mg/L, pH = 7.5), suspended solid (SS) concentration and turbidity of coal washing wastewater treated by TPADM reached 14.3 mg/L and 8.7 NTU, respectively. The floc formation, breakage, and regeneration analyses showed that the cationic fragment structure in TPADM could contribute to the formation of large and high density flocculation structure (d(50) >= 245 mu m,D-f >= 2.05), and these coal flocs can regenerate rapidly after being broken. Finally, these large and compact flocs help to accelerate the flocs sedimentation and reduce the SS concentration and turbidity of coal washing wastewater, and enhance the effect of solid-liquid separation.

The combination of applying KCl instead of NaCl together with phosphorous limitation significantly increased P(3HB) productivity to 0.25 g L-1 h(-1) compared to 0.09 g L-1 h(-1). It can be concluded that the effective utilization of sucrose as a carbon source requires a combination of high nitrogen and low phosphorous concentration and a salt content of 0.6 g L-1 KCl for P(3HB) production with B. megaterium uyuni S29.

Backgrounds Pomegranate (Punica granatum L.) is an important commercial fruit tree, with moderate tolerance to salinity. The balance of Cl- and other anions in pomegranate tissues are affected by salinity, however, the accumulation patterns of anions are poorly understood. The chloride channel (CLC) gene family is involved in conducting Cl-, NO3-, HCO3- and I-, but its characteristics have not been reported on pomegranate. Results In this study, we identified seven PgCLC genes, consisting of four antiporters and three channels, based on the presence of the gating glutamate (E) and the proton glutamate (E). Phylogenetic analysis revealed that seven PgCLCs were divided into two clades, with clade I containing the typical conserved regions GxGIPE (I), GKxGPxxH (II) and PxxGxLF (III), whereas clade II not. Multiple sequence alignment revealed that PgCLC-B had a P [proline, Pro] residue in region I, which was suspected to be a NO3-/H+ exchanger, while PgCLC-C1, PgCLC-C2, PgCLC-D and PgCLC-G contained a S [serine, Ser] residue, with a high affinity to Cl-. We determined the content of Cl-, NO3-, H2PO4-, and SO42- in pomegranate tissues after 18 days of salt treatments (0, 100, 200 and 300 mM NaCl). Compared with control, the Cl- content increased sharply in pomegranate tissues. Salinity inhibited the uptake of NO3- and SO42-, but accelerated H2PO4- uptake. The results of real-time reverse transcription PCR (qRT-PCR) revealed that PgCLC genes had tissue-specific expression patterns. The high expression levels of three antiporters PgCLC-C1, PgCLC-C2 and PgCLC-D in leaves might be contributed to sequestrating Cl- into the vacuoles. However, the low expression levels of PgCLCs in roots might be associated with the exclusion of Cl- from root cells. Also, the up-regulated PgCLC-B in leaves indicated that more NO3- was transported into leaves to mitigate the nitrogen deficiency. Conclusions Our findings suggested that the PgCLC genes played important roles in balancing of Cl- and NO3- in pomegranate tissues under salt stress. This study established a theoretical foundation for the further functional characterization of the CLC genes in pomegranate.

The soliton mode-locked laser operated at 1563 nm with the shortest pulse width of 950 fs and thus it could be applied in optical communications.

Background: International guidelines recommend either intravenous immunoglobulin (IVIg) or corticosteroids as first-line treatment for chronic inflammatory demyelinating polyradiculoneuropathy (CIDP). Corticosteroids act more slowly and are associated with more side effects, but may induce long-term remissions. The hypothesis of this study is that combined IVIg and corticosteroid induction treatment will lead to more frequent long-term remissions than IVIg treatment alone. Methods: An international, randomised, double-blind, placebo-controlled trial, in adults with ‘probable’ or ‘definite’ CIDP according to the EFNS/PNS 2010 criteria. Three groups of patients are included: (1) treatment naive, (2) known CIDP patients with a relapse after > 1 year without treatment, and (3) patients with CIDP who improved within 3 months after a single course of IVIg, who subsequently deteriorate at any interval without having received additional treatment. Patients are randomised to receive 7 courses of IVIg and 1000 mg intravenous methylprednisolone (IVMP) (in sodium chloride 0.9%) or IVIg and placebo (sodium chloride 0.9%), every 3 weeks for 18 weeks. IVIg treatment consists of a loading dose of 2 g/kg (over 3-5 days) followed by 6 courses of IVIg 1/g/kg (over 1-2 days). The primary outcome is remission at 1 year, defined as improvement in disability from baseline, sustained between week 18 and week 52 without further treatment. Secondary outcomes include changes in disability, impairment, pain, fatigue, quality of life, care use and costs and (long-term) safety. Discussion: In case of superiority of the combined treatment, patients will experience the advantages of two proven efficacious treatments, namely rapid improvement due to IVIg and long-term remission due to corticosteroids. Long-term remission would reduce the need for maintenance IVIg treatment and may decrease health care costs. Additionally, we expect that the combined treatment leads to a higher proportion of patients with improvement as some patients who do not respond to IVIg will respond to corticosteroids. Meanwhile, the fluorescence intensity of Cu-CDs decreased gradually along with the increasing of pH and a sigmoidal curve was well fitted with the pK(a1)2.49 and pK(a2) 6.21, respectively. The Cu-CDs as a multi-functional fluorescence sensing platform demonstrated great accuracy and selectivity for tetracyclines (TCs) and pH. On account of practical application, as-synthesized Cu-CDs not only were effectively employed for determination of TCs in antibiotic tablets and milk products, but also could trace nuclear and had been extended to sense and visualize pH fluctuations in cells due to low toxicity and biocompatibility. As a facile and effective multi-functional fluorescent sensor, the proposed Cu-CDs have a significant potential for biosensing and biomedical imaging.
