A series of 1-(substituted benzyl)-4-(5-(5-nitroheteroaryl-2-yl)-1,3,4-thiadiazol-2-yl) piperazine derivatives I [X = O, S; R= 3-MeO, 2-O2N, 2,5-(Cl)2, 2,4,5-(F)3, etc.] were synthesized. The synthesized compounds I were evaluated against three metronidazole-resistant isolates of Helicobacter pylori using disk diffusion bioassay test. The results of SAR study indicated that the potency and anti-H. pylori activity profile of synthesized derivatives was mainly attributed to the substituted nitroaryl moiety at the C-5 position of 1,3,4-thiadiazole ring. Compound I [X = O; R = 3-MeO] was found to be the most potent.
