A methylene violet 3RAX-conjugated porphyrin has been designed and synthesized as a potential photosensitizer in photodynamic therapy because of its high DNA binding constant and the efficacy in inhibiting cancer cells with subcellular localization. At 400μM, the DNA photocleavage activities of compound 1 and2 were 80% and 20%, resp. The results fromphotophys. studies and cell experiments suggest porphyrin-methylene violet 3RAX conjugate could be developed as potential photosensitizers for mitochondrial targeting action in PDT.
