The cucurbit[8]uril (CB8) synthetic receptor was shown to form high-affinity host-guest complexes with dicationic dithienylethene (DTE) photoswitches I [R = H, Me; R1 = 1-iodo-1-methyl-2-pyridyl, 1-iodo-1-methyl-3-pyridyl, 1-iodo-1-methyl-4-pyridyl] in water. ITC experiments combined with computational studies suggest that the formation of the inclusion complexes was mainly driven by a combination of hydrophobic effects, ion-dipole, hydrogen- and chalcogen-bonding interactions. The binding affinities were observed to be much higher for the DTE closed isomers, reaching values in the picomolar range (up to 1011 M-1) while the open isomers display up to 10 000-fold lower affinities, setting ideal conditions for the development of robust photoswitchable host-guest complexes. The light-responsive affinity of these photoswitches toward CB8 was explored to control the encapsulation and release of nanomolar affinity steroids via competitive guest replacement.

It is successfully demonstrated that the ability to modulate the luminescence from upconverting nanoparticles in vitro and in vivo by taking advantage of the well-understood photochem. of DTE mol. switches decorated onto nanoparticle surfaces by the use of CuACC chem. By reversibly converting the photoswitch back and forth between the two isomers by using UV and visible light, we are able to toggle the system between on (bright fluorescence) and off (quenched fluorescence) states. This process can be cycled multiple times before any degradation of the nanoparticles or the photo-switch renders the system impractical. The system has an advantage over ‘caged’ fluorophores as the luminescence of our designed nanoparticles can be repeatedly cycled through on and off states. Systems such as the one described not only have the capacity to reversibly control the intensity of the probe emission, through selective absorption, they can also alter the color of the emitted light from multicolored probes; this further enhances the imaging capabilities.
