In this study, we monitored culturable and resistant wastewater bacteria and cross-resistance to clinically relevant antibiotics to gauge the impact of each antimicrobial and identify factors influencing cross-resistance profiles. Bacteria resistant to TCS and BC were isolated from wastewater influent over 21 months, and cross-resistance, taxonomy, and monthly changes were characterized under both antimicrobial selection regimes. Cross-resistance profiles from each antimicrobial differed within and between taxa. BC-isolated bacteria had a significantly higher prevalence of resistance to last-resort antibiotic colistin, while isolates resistant to TCS exhibited higher rates of multidrug resistance. Prevalence of culturable TCS-resistant bacteria decreased over time following Food and Drug Administration (FDA) TCS bans. Cross-resistance patterns varied according to sampling date, including among the most clinically important antibiotics. Correlations between strain-specific resistance profiles were largely influenced by taxonomy, with some variations associated with sampling date. The results reveal that time, taxonomy, and selection by TCS and BC impact features of cross-resistance patterns among diverse wastewater microorganisms, which could reflect the variety of factors influencing resistance patterns relevant to a community microbiome.

Cataracts have a high incidence and prevalence rate worldwide, and they are the leading cause of blindness. Lens epithelial cell (LEC) apoptosis is often analysed in cataract research since it is the pathological basis of cataracts, except for congenital cataract. Chloride channels are present in ocular tissues, such as in trabecular cells, LECs and other cells. They serve an important role in apoptosis and participate in endoplasmic reticulum (ER) stress and oxidative stress. However, their role in the apoptosis of LECs has not been discussed. The present study examined the effects of the chloride channel blocker 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) in human LECs (HLECs) to elucidate the role of NPPB in HLECs and investigate the role and mechanism of chloride channels in cataract formation. HLECs were exposed to NPPB. Cell survival rate was evaluated using Cell Counting Kit-8 assays. Oxidative stress was detected as reactive oxygen species (ROS) in cells by using a ROS assay kit. Apoptosis was examined by assessing mitochondrial membrane potential and using a JC-1 assay kit, and western blot analysis was performed to measure the expression levels of mitochondrial-dependent apoptosis pathway-associated proteins. ER stress was evaluated by determining the intracellular calcium ion fluorescence intensity, and western blot analysis was performed to measure ER stress-associated protein expression. The results revealed that NPPB treatment decreased the viability of HLECs and increased apoptosis. Additionally, NPPB increased intracellular ROS levels, as well as the number of JC-1 monomers and the protein expression levels of B-cell lymphoma-2 (Bcl-2)-associated X and cleaved caspase-3, and decreased Bcl-2 protein expression. NPPB increased intracellular calcium ions, the protein expression levels of activating transcription factor 6, JNK, C/EBP homologous protein and caspase-12, and the phosphorylation of protein kinase R-like endoplasmic reticulum kinase. N-acetylcysteine and 4-phenylbutyric acid inhibited NPPB-induced oxidative stress, ER stress and apoptosis. Therefore, NPPB treatment decreased cell viability and promoted apoptosis of HLECs via the promotion of oxidative and ER stress.

Corrosion induced failure of buried ferrous pipelines causes significant economic losses to the world. Many of the existing buried water pipelines are made of ductile iron in most of the developed countries. Literature shows that the research on the corrosion-induced degradation assessments of ductile iron pipelines is limited and mostly deals with the physical deterioration i. e., loss of wall thickness determined by short-term research. This paper addresses this gap and presents a comparatively long-term study on the corrosion and its subsequent effect on the composition and nanomechanical properties of buried ductile iron pipelines in the accelerated corrosive environment. For experimentation, ductile iron specimens were immersed in the acidic simulated soil solution for 365 days. Physical deterioration assessment over the time was carried out in terms of corrosion rates measured at intervals of 180 and 365 days. A significant change in the percentage composition of key elements and phases were quantified. Furthermore, the mechanical properties of the grains were found to be considerably reduced after 365 days of immersion in the acidic environment by the nanoindentation technique. The results revealed an alarming increase of structural degradation at nano scale for some of the grains due to chloride induced localised corrosion. The significance of the current research is its in-depth analysis of the corrosion-induced degradation of ductile iron pipes which enhances the knowledge related to the failure of these pipelines.

Influence of typical electrolytes on electrooxidation of bio-refractory reactive dye

Electrochemical oxidation is a promising alternative for the degradation of reactive dyestuffs in residual dyeing liquid, which contains organic dyes, as well as salts such as sodium chloride and sodium sulfate. In this work, three kinds of salts, Na2SO4, NaCl and FeSO4, were selected to study the influence of electrolytes on the electrochemical oxidation of recalcitrant reactive red X-3B, using graphite cathode and dimensionally stable anode or graphite anode. The removals of color and chemical oxygen demand have been evaluated. Color removals were 99.97% and 99.84% with graphite anode and dimensionally stable anode in Na2SO4 and NaCl electrolyte. The corresponding chemical oxygen demand removals were 93.60% and 100%. In contrast, the maximum color and chemical oxygen demand removals were 97.32% and 68.76% in FeSO4 electrolyte. It was found that a combination of NaCl and FeSO4 achieved complete oxygen demand removal for both graphite anode and dimensionally stable anode. During the electrochemical oxidation process, anode and electrolyte exhibited a great effect on pH variations. Based on the cyclic voltammetry and active oxidative species investigations, the oxidation of reactive red X-3B was attributed to indirect oxidation of active chlorine and reactive oxygen species.

Developed Process Circuit Flowsheet of Al Amar Ore for Production of Nanocrystalline Ferrite and Improving Gold Recovery

Al Amar gold ore is rich in sulfides of base metals and is commercially applied for the production of copper concentrate via floatation and gold bullion by cyanidation of tailing. The current process flowsheet suffers from low gold recovery (similar to 60%) and loss of metals in the hazardous stockpiled residue. This work addresses these drawbacks by a newly experimental redesign of the process circuit. The innovative flowsheet comprises a sequence of operations, including acid leaching of the roasted ore, gold recovery from the leach residue, and preparation of a valuable zinc-copper-lead ferrite from the filtrate by coprecipitation followed by heat treatment. The ore is roasted at 650 degrees C and then leached in 20% HCI, where most of Zn, Cu, Pb, and Fe contents are dissolved, while pristine gold remains in the residue. Most of the gold (similar to 93%) can be recovered by cyanidation of the acid leach residue. Stoichiometric ratios of dissolved Zn, Cu, Pb, and Fe in the acid leach solution can be kept at 0.6:0.3:0.1:2.0, respectively, only by adding a small amount of ferric chloride. These metals are coprecipitated at varying pH values from 8 to 10, and the produced powders are annealed at temperatures from 600 to 1100 degrees C. X-ray diffraction (XRD) charts reveal sharp peaks of the targeted Zn0.6Cu0.3Pb0.1Fe2O4 phase at 600 degrees C, while a highly crystalline single phase is obtained at 1100 degrees C, independently of precipitation pH. The crystalline size of the produced powders increases with annealing temperatures (from 18-27 nm at 600 degrees C to 85-105 nm at 1100 degrees C). The finest size is found at pH 12. Scanning electron microscopy (SEM) investigation shows uniform cubic microstructures of samples annealed at 1100 degrees C. The produced ferrite powders exhibit soft magnetic characteristics. Saturation magnetization, M-s, substantially increases with pH. Coercivity, H-c, increases with increasing annealing temperatures, from 600 to 800 degrees C, and decreases above 800 degrees C. Preliminary cost-benefit analysis revealed that the profit margin of the proposed process flowsheet is promising. The wastewater is almost free of heavy metals. Our advances in high gold recovery and preparation of valuable magnetic nanocrystalline ferrite provide exciting opportunities to enhance and maximize Al Amar ore production for practical applications.

Objective This study aimed to develop a nomogram to predict the neck occult metastasis in early (T1-T2 cN0) oral squamous cell carcinoma (OSCC). Materials and Methods The nomogram was developed in a training cohort of 336 early OSCC patients and was validated in a validation cohort including 88 patients. Independent predictors were calculated by univariate and multivariate logistic regression analyses. Results In univariate logistical regression analysis, gender, perineural invasion (PNI), blood vessel invasion, mean corpuscular hemoglobin, aspartate aminotransferase, prealbumin, globulin (GLO), lactate dehydrogenase (LDH), serum sodium (NA), and serum chloride were significant associated with neck occult metastasis. Multivariate logistical regression analysis identified PNI (p < .001), LDH (p = .003), GLO (p = .019), and NA (p = .020) as independent predictors of neck occult metastasis. Cut-off values for LDH, GLO, and NA obtained from AUC were 142.5, 26.35, and 139.5, respectively. The nomogram based on PNI and categorical GLO, LDH, and NA exhibited a strong discrimination, with a C-indexes of 0.748 (95%CI = 0.688 to 0.810) in the training cohort and 0.751 (95%CI = 0.639 to 0.863) in the validation cohort. Conclusions A nomogram based on PNI, LDH, GLO, and NA for predicting the risk of neck lymph nodes occult metastasis in OSCC could help surgeons with therapy decision-making. Evaluation of antioxidant and anticancer activity of crude extract and different fractions of Chlorella vulgaris axenic culture grown under various concentrations of copper ions

Background Chlorella vulgaris is a microalga potentially used for pharmaceutical, animal feed, food supplement, aquaculture and cosmetics. The current study aims to study the antioxidant and prooxidant effect of Chlorella vulgaris cultivated under various conc. of copper ions. Methods The axenic green microalgal culture of Chlorella vulgaris was subjected to copper stress conditions (0.00, 0.079, 0.158, 0.316 and 0.632 mg/L). The growth rate was measured at OD680 nm and by dry weight (DW). Moreover, the Antioxidant activity against DPPH and ABTS radical, pigments and phytochemical compounds of the crude extracts (methylene chloride: Methanol, 1:1) were evaluated. The promising Cu crude extract (0.316 mg/L) further fractionated into twenty-one fractions by silica gel column chromatography using hexane, chloroform and ethyl acetate as a mobile phase. Results The obtained results reported that nine out of these fractions exhibited more than 50% antioxidant activity and anticancer activity against Hela cancer cell lines. Based on IC50, fraction No. 7 was found to be the most effective fraction possessing a significant increase in both antioxidant and anticancer potency. Separation of active compound (s) in fraction No 7 was performed using precoated silica gel plates (TLC F-254) with ethyl acetate: hexane (9:1 v/v) as mobile phase. Confirmation of active compound separation was achieved by two-dimensional TLC and visualization of the separated compound by UV lamp. The complete identification of the separated active compound was performed by UV- Vis- spectrophotometric absorption, IR, MS, H-1-NMRT C-13-NMR. The isolated compound ((2E,7R,11R)-3,7,11,15-Tetramethyl-2-hexadecenol) have high antioxidant activity with IC50 (10.59 mu g/ml) against DPPH radical assay and comparable to the capacities of the positive controls, Butylated hydroxy toluene [BHT] (IC50 11.2 mu g/ml) and Vitamin C (IC50 12.9 mu g/ml). Furthermore, pure isolated compound exhibited a potent anticancer activity against Hela cell line with IC50 (4.38 mu g/ml) compared to Doxorubicin (DOX) as synthetic drug (13.3 mu g/ml). In addition, the interaction of the pure compound with Hela cancer cell line and gene expression were evaluated.
