Several guests have been encapsulated in a dendritic box. The maximum number of guests is determined by the shape and size of cavities and guests. Shape-selective liberation of guests with different sizes is achieved by the hydrolysis of the shell in a two-step process. First the shell is perforated by the hydrolysis of the protecting N-t-BOC groups of the L-Phe end group, furnishing the liberation of small guests (p-nitrophenol, p-nitro-benzoic acid or an EPR probe) upon dialysis. The larger guests–bengal rose, rhodamine, new coccine and methylene violet 3RAX–still being imprisoned in the box with a perforated shell, are liberated by subsequent hydrolysis of the amide groups. By changing the amino acid and the protected groups used, it is possible to tune the shape-selectivity in the liberation of guests of different size.

Organic dyes can be excellent catalysts for photoredox chem., offering low price, low toxicity, and an exceptional range of available materials. Their use has been limited because in comparison to their transition-metal catalysts the spectroscopic, kinetic, and electrochem. information available is far more limited. To remediate this situation, we have determined the necessary data for 14 readily available dyes with excellent potential as photoredox catalysts. We have also demonstrated the utility of these dyes through visible-light-mediated reductive dehalogenation and Aza-Henry reactions. We envision that this collection of data will lead to an increase in the use of cationic dyes in photoredox processes because users will find the necessary information readily available.

The authors describe the efficient synthesis and one-step derivatization of novel, nonfluorescent azo dyes based on the Black Hole Quencher-3 (BHQ-3) scaffold. These dyes were equipped with various reactive and/or bioconjugatable groups (azido, α-iodoacetyl, ketone, terminal alkyne, vicinal diol). The azido derivative is highly reactive in the context of copper-catalyzed azide-alkyne cycloaddition (CuAAC) reactions and allowed easy synthetic access to the first water-soluble (sulfonated derivative) and aldehyde-modified BHQ-3 dyes, the direct preparation of which failed by conventional azo-coupling reactions. The aldehyde- and α-iodoacetyl-containing fluorescence quenchers were readily conjugated to aminooxy- and cysteine-containing peptides by the formation of a stable oxime or thioether linkage, resp. Further fluorescent labeling of the resultant peptide conjugates with red- or far-red-emitting rhodamine or cyanine dyes through sequential and/or one-pot bioconjugations, led to novel Foerster resonance energy transfer (FRET) based probes suitable for the in vivo detection and imaging of urokinase plasminogen activator, a key protease in cancer invasion and metastasis.

In 1891, P. Guttmann and P. Ehrlich (1891) were the first to report the antimalarial properties of a synthetic, rather than a natural, material when they described the clin. cure of two patients after oral administration of a thiazine dye, methylene blue. Since that time, sporadic reports of the antimalarial properties of several xanthene and azine dyes related to methylene blue have been noted. We report here the results from a reexamination of the antimalarial properties of methylene blue, Janus green B, and three rhodamine dyes and disclose new antimalarial data for 16 com. available structural analogs of these dyes. The 50% inhibitory concentrations for the chloroquine-susceptible D6 clone and SN isolate and the chloroquine-resistant W2 clone of Plasmodium falciparum were determined by the recently described parasite lactate dehydrogenase enzyme assay. No cross-resistance to chloroquine was observed for any of the dyes. For the 21 dyes tested, no correlation was observed between antimalarial activity and cytotoxicity against KB cells. No correlation between log P (where P is the octanol/water partition coefficient) or relative catalyst efficiency for glucose oxidation and antimalarial activity or cytotoxicity was observed for the dyes as a whole or for the thiazine dyes. Thiazine dyes were the most uniformly potent structural class tested, and among the dyes in this class, methylene blue was notable for both its high antimalarial potency and selectivity.

Microvia filling by Cu electroplating was carried out using a plating bath containing a suppressor and a leveler without an accelerator. The required accelerator was adsorbed onto the Cu seed layer of the microvia in a predipping bath before the filling plating. This pretreatment is similar to the self-assembled monolayer of thiol mols. that forms on a Cu surface. The suppressor was poly(ethylene glycol) (PEG), and 5 levelers, namely, Janus green B, diazine black, methylene violet, safranine O, and Alcian blue, were employed to screen for the best plating formula suitable for a plating process in which no accelerator was present in the plating solution The thiol mol. employed in this work was 3-mercapto-1-propanesulfonate (MPS). The electrochem. behaviors of various plating formulas were characterized using a galvanostatic measurement on a Cu electrode at different rotating speeds. The MPS adlayer is transferable onto the surface of the Cu deposit and can be displaced by the PEG-Cl-leveler. The displacement rate depends on the mol. structure of the added leveler. This plating process has the potential to greatly reduce the plating time of microvia filling.

The structural and energetic aspects of the interaction of two important phenazinium dyes methylene violet 3RAX and indoine blue with DNA were studied by spectroscopic and thermochem. techniques. The binding affinity values of both the dyes were of the order of 105 M-1 but methylene violet 3RAX bound to DNA more strongly. Both dyes bound DNA by intercalation but methylene violet 3RAX bound stronger than indoine blue. The DNA binding of both the dyes was exothermic and favored by a small neg. enthalpy and pos. entropy contributions. The effect of ionic strength indicated that electrostatic attraction is an important component of the dye-DNA interaction although the major contribution comes from hydrophobic forces. The temperature dependence of enthalpy changes yielded neg. heat capacity value which was higher for methylene violet 3RAX compared to indoine blue suggesting higher contribution from hydrophobic forces in the interaction of the former. Enthalpy-entropy compensation phenomenon was observed for the binding of both dyes to DNA.

Industrial effluents with dyes may contain appreciable concentrations of materials with high COD and suspended solids, posing adverse effects to both humans and aquatic life; therefore, these effluents require quant. monitoring. In the present study, an anal. method based on solid-phase extraction (SPE) and ultra-performance liquid chromatog.-tandem mass spectrometry (UPLC-MS/MS) has been optimized for the quant. anal. of methylene blue (MB) in environmental samples. For the extraction of MB, a variety of solvents, including formic acid, were investigated to obtain optimum recovery. MB extraction using the SPE method was best achieved using methanol with 1 M formic acid. Chromatog. separation of MB and methylene violet 3RAX (MV 3RAX, internal standard) was accomplished on an Acquity BEH C18 column using water (64.99%) with formic acid (0.01%) and acetonitrile (35%) in isocratic elution mode. Chromatog. separation for both MB and MV 3RAX was achieved in <2 min with good resolution and superior peak symmetry. MB and MV 3RAX were quantified with electrospray ionization coupled with MS/MS in the multiple reaction monitoring mode. Good quality parameters were achieved for instance linearity (r2 > 0.999), run-to-run and day-to-day precisions with relative standard deviations of <4%, sensitivity with a low limit of detection (LOD) of 0.1 ng mL-1 and limit of quantification (LOQ) of 0.4 ng mL-1 were obtained. Industrial wastewater samples (paper, textile, laundry and printing press) were analyzed, and an MB level was found in between 0.36 and 1.08 μg mL-1 with excellent recovery rates (95-99%) depending on samples.
