It was found that decamethoxin, as a broad-spectrum antiseptic with a pronounced antibacterial and fungicidal action, and BF-6 phenol-polyvinylacetal adhesive, which gives complex sutures the structure of a pseudomonofilament, are best as antimicrobial additives in a modifying composite for giving surgical sutures antimicrobial properties. A decamethoxin content in the modifying composite of 10% of the weight of BF-6 (1.7-2.0% in the suture) is sufficient for manifestation of the antimicrobial effect with respect to Staphylococcus aureus, E. coli, and yeast-like fungi. The antimicrobial activity of a suture with a 1.7-2.0 weight % decamethoxin content in the period from 2 to 24 h increases by 4 times with respect to E. coli and by 8 times with respect to Staphylococcus aureus and yeast-like fungi. Application of a coating of BF-6 adhesive with decamethoxin has almost no effect on the physicomech. properties of the sutures, but the capillarity decreases significantly (by ∼86%), which also decreases the probability of wound infection. The kinetics of release of decamethoxin does not change after radiation sterilization of the sutures, i.e., the composition and content of the antimicrobial additive in the modified sutures and its antimicrobial activity are preserved. The optimum medium for storage of sterile modified sutures is 1% decamethoxin solution After 12 days of storage, the sutures remain sterile and lose a maximum of 6% of the breaking strength.

Effects caused by combined inclusion of two membranotropic agents (MTA) of different nature into model phospholipid membranes have been examined An effect of significant change in activity of individual M1IA based on quaternary and bisquaternary ammonium compounds (QAC, BQAC) – decamethoxinum. aethonium. tetramethylammonium – and organic 2.5-dihydroxybcnzoic acid (DHB) on their combined incorporation into model multibilayer structures of hydrated dipalmitoylphosphatidycholine (DPPC) is revealed. Differential scanning calorimetry (DSC) data have shown that, when both types of MTA are jointly present in the system. significant deviations from additivity values are noted for the measured calorimetry parameters. This shows that it is the MTA complexes. rather than individual MTAs. affect the model membrane properties. Mass-spectrometric and quantum chem. calculations data have demonstrated a possibility of formation of stable noncovalent complexes including (di)cations of quaternary ammonium bases and anions of organic DHB acid. Plotting of “membrane melting temperature (Tm) vs. MTA composition” quasi-binary diagrams permitted us to determine the optimal stoichiometry of the BQAC-DHB membrane-incorporated complexes. It was revealed by means of DSC that incorporation into membranes of individual ionic BQAC and DHB as well as their complexes with stoichiometry providing preservation of the ionic (charged) state of the MTAs causes decrease of Tm and correspondingly, disordering of the membrane structure. At the same time, under MTA concentrations providing neutral complexes of BQAC and DHB. some increase in Tm is observed corresponding to an ordering of membrane structure. which can be interpreted as deactivation of BQAC. i.e. modulation of their activity in the presence of the organic acid. The ascertained possibility of suppression of membranotropic activity of antimicrobial BQAC on their combined use with an organic acid is to be taken into account in design of multi-component antimicrobial drugs.

A model mol. biosystem of hydrated dipalmitoylphosphatidylcholine (DPPC) bilayers that mimics cell biomembranes is used to probe combined membranotropic effects of drugs by instrumental techniques of mol. biophysics. Differential scanning calorimetry reveals that doping of the DPPC model membrane with individual bisquaternary ammonium compounds (BQAC) decamethoxinum, ethonium, thionium and acetylsalicylic acid (ASA) leads to lowering of the membrane melting temperature (Tm) pointing to membrane fluidization. Combined application of the basic BQAC and acidic ASA causes an opposite effect on Tm (increase), corresponding to the membrane densification. Thus, modulation of the membranotropic effects upon combined use of the drugs studied can be revealed at the level of model membranes. Formation of noncovalent supramol. complexes of the individual BQACs and ASA with DPPC mols., which may be involved in the mechanism of the drug-membrane interaction at the mol. level, is demonstrated by electrospray ionization (ESI) mass spectrometry. In the ternary (DPPC + ASA + BQAC) model systems, the stable complexes of the BQAC dication with the ASA anion, which may be responsible for modulation of the membranotropic effects of the drugs, were recorded by ESI mass spectrometry. The proposed approach can be further developed for preliminary evaluation of the combined effects of the drugs at the level of model lipid membranes prior to tests on living organisms.
